It\u2019s as easy as wearing a specialized set of goggles and a headset once a day for an hour. Does this sound too good to be true? The <a href=\"https:\/\/www.alz.org\/\" target=\"_blank\" rel=\"noopener\">Alzheimer\u2019s Association<\/a> states that 55 million people worldwide live with Alzheimer&#8217;s and other dementias.<\/p>\n<blockquote><p>Alzheimer\u2019s disease is a degenerative brain disease and the most common form of dementia. Dementia is not a specific disease. It&#8217;s an overall term that describes a group of symptoms. \u2013 The Alzheimer\u2019s Association<\/p><\/blockquote>\n<p>Dementia is a devastating disease for patients and their families. Beyond the suffering, a high societal cost is associated with caring for dementia patients. According to <em><a href=\"https:\/\/www.ajmc.com\/view\/the-economic-and-societal-burden-of-alzheimer-disease-managed-care-considerations\" target=\"_blank\" rel=\"noopener\">The American Journal of Managed Care<\/a><\/em>, \u201cIn 2022, the estimated healthcare costs associated with [Alzheimer\u2019s disease] treatment were $321 billion, with costs projected to exceed 1 trillion [dollars] by 2050. These cost-of-care projections are based on direct healthcare costs and are likely underestimated because indirect costs associated with AD treatment are usually not included.\u201d<\/p>\n<p>As the world ages, there is an increased need for treatments for Alzheimer&#8217;s disease.<\/p>\n<p>Cognito Therapeutics was founded in 2016 after Professors Ed Boyden and Li-Huei Tsai, both of MIT, discovered how to trigger gamma waves in mice. The gamma waves reduced the amyloid plaques in the mice\u2019s brains. Since amyloid plaques accumulate in Alzheimer&#8217;s patients, they focused on applying gamma wave stimulation to human patients.<\/p>\n<h2>Gamma Wave Therapy<\/h2>\n<p>The technique, known as gamma wave therapy, uses a headset that delivers pulsing lights and sounds at fast and specific frequencies. The therapy aims to stimulate the activity of immune cells inside the brain known as microglia. The aim is to help the central nervous system clear out specific proteins that can lead to neurological diseases and dementia.<\/p>\n<p>In the study <a href=\"https:\/\/journals.plos.org\/plosone\/article?id=10.1371\/journal.pone.0278412\" target=\"_blank\" rel=\"noopener\">published in PLOS ONE<\/a> in December 2022 that showed that six months\u2019 use of their digital therapeutic could reduce the rate of atrophy within the brain&#8217;s white matter, the researchers from Cognito Therapeutics used <a href=\"https:\/\/www.mathworks.com\/matlabcentral\/fileexchange\/56415-eeglab?s_tid=srchtitle_site_search_1_eeglab\" target=\"_blank\" rel=\"noopener\">EEGLAB<\/a>, a MATLAB community toolbox for processing electrophysiological signals.<\/p>\n<p>&nbsp;<\/p>\n<p><div id=\"attachment_3815\" style=\"width: 410px\" class=\"wp-caption alignnone\"><a href=\"https:\/\/journals.plos.org\/plosone\/article?id=10.1371\/journal.pone.0278412\" target=\"_blank\" rel=\"noopener\"><img aria-describedby=\"caption-attachment-3815\" decoding=\"async\" loading=\"lazy\" class=\"wp-image-3815\" src=\"https:\/\/blogs.mathworks.com\/headlines\/files\/2023\/08\/journal.pone_.0278412.g003-.png\" alt=\"\" width=\"400\" height=\"424\" \/><\/a><p id=\"caption-attachment-3815\" class=\"wp-caption-text\">Acute 40Hz combined visual and auditory stimulation entrains cortical and subcortical regions. Image credit: Chan D, Suk H-J, Jackson BL, Milman NP, Stark D, Klerman EB, et al.<\/p><\/div><\/p>\n<p>&nbsp;<\/p>\n<p>In the same study, the researchers at Cognito Therapeutics used Statistical Parametric Mapping (<a href=\"https:\/\/www.mathworks.com\/matlabcentral\/fileexchange\/68729-statistical-parametric-mapping?s_tid=srchtitle_site_search_2_spm\">SPM<\/a>), a MATLAB Community Toolbox, \u00a0for the MRI analysis of the study participants. The SPM software package has been designed to analyze brain imaging data sequences. The sequences can be a series of images from different cohorts, or time series from the same subject. The current release is designed to analyze <a href=\"https:\/\/www.fil.ion.ucl.ac.uk\/spm\/doc\/biblio\/Keyword\/FMRI.html\" target=\"_blank\" rel=\"noopener\">fMRI<\/a>,\u00a0<a href=\"https:\/\/www.fil.ion.ucl.ac.uk\/spm\/doc\/biblio\/Keyword\/PET.html\" target=\"_blank\" rel=\"noopener\">PET<\/a>,\u00a0<a href=\"https:\/\/www.fil.ion.ucl.ac.uk\/spm\/doc\/biblio\/Keyword\/PET.html\" target=\"_blank\" rel=\"noopener\">SPECT<\/a>,\u00a0<a href=\"https:\/\/www.fil.ion.ucl.ac.uk\/spm\/doc\/biblio\/Keyword\/EEG.html\" target=\"_blank\" rel=\"noopener\">EEG<\/a>\u00a0and\u00a0<a href=\"https:\/\/www.fil.ion.ucl.ac.uk\/spm\/doc\/biblio\/Keyword\/MEG.html\" target=\"_blank\" rel=\"noopener\">MEG<\/a>.<\/p>\n<p>The anatomical changes are shown in the figure below. However, it was limited in scope due to restrictions during the pandemic. Cognito Therapeutics has <a href=\"https:\/\/www.hopestudyforad.com\/\" target=\"_blank\" rel=\"noopener\">started a much larger clinical trial<\/a> which will enroll approximately 500 patients at 50 sites. This phase three test will focus on patients with mild to moderate disease.
